To develop a HPLC method for the determination of 1,3,6-trihydroxy-2-methylanthra-quinone-3-O-[3-O-acetyl--L-rhamnopyranosyl-(12)--D-glucopyranoside] in Rubia.cordifolia for the first time.

HPLC was carried out on a Purospher star RP C18 column(250 mm 4.6 mm,5 m) at 25 using MeOH-H2O-tetrahydropyrane (65:34.7:0.3) as mobile phase,at a flow rate of 1.0 mL/min.The detection wavelength was 276 nm.

The linearity was found in the range of 0.020-0.160 g,r=0.9998,the average recovery was 101.5%,RSD was 2.0%.This method is reliable,accurate and suitable for the determination of 1,3,6-trihydroxy-2-methylanthraquinone-3-O-[3-O-acetyl--L-rhamnopyranosyl-(12)--D-glucopyranoside].
